The first permanent European settlers arrived in 1653 from New Sweden, and the area eventually came under English control in 1664, being included in a large land grant to William Penn in 1681. Upon the founding of the colony of Pennsylvania, the area became part of Darby Township in Chester County, which later formed part of Delaware County in 1789.

One notable historical site in Upper Darby is the Lower Swedish Cabin, built around 1654 and considered a remnant of the early Swedish settlers who introduced the log cabin to the area. The township was also active in the antislavery movement, with many homes serving as stops on the Underground Railroad.

The 19th century brought significant industrialization to Upper Darby, which led to a population increase and the development of various industries, including mills, rock quarries, factories, and other industrial concerns. This industrial growth attracted a wave of European immigrants and numerous churches were founded. This farm is a 19th-century historical farmhouse and barn. The farm was added to the National Register of Historic Places in 1988 and now functions as a museum.

Upper Darby has been home to some famous personalities over the years. Actress and comedian Tina Fey grew up in Upper Darby. Others who have resided in the area include composer Mark lburger and Olympic rower Daniel Lyons. Some scenes from the movie Silver Linings Playbook (2011) were recorded in Upper Darby as well.
